What Is GMP and Why Is It Mandatory?
Good Manufacturing Practice (GMP) is an internationally recognised standard that ensures pharmaceutical and medical products are consistently produced and controlled according to quality standards. In Turkey, the Ministry of Health, and in EU countries, the EMA (European Medicines Agency) enforce compliance with these standards.
A production line without GMP certification cannot release products to market. Therefore, machine selection in the pharmaceutical sector is a specialist discipline in its own right.
5 Critical Factors in Line Design
1. Cleanroom Classification (ISO 5 / Grade A)
For injectable products, cleanroom quality is the most critical parameter. According to ISO 14644:
- ISO 5 (Grade A): Filling/closing zone — particle count ≤3,520/m³ (≥0.5 µm)
- ISO 7 (Grade C): Preparation areas
- ISO 8 (Grade D): General production corridors
Aseptic filling machines must be equipped with RABS (Restricted Access Barrier Systems) or isolator technology.
2. CIP/SIP Integration
Clean-in-Place (CIP) and Sterilise-in-Place (SIP) systems enable decontamination without stopping the line. For modern pharmaceutical lines this is mandatory; both GMP Annex 1 and FDA 21 CFR Part 211 explicitly define this requirement.
3. Validation Packages (IQ / OQ / PQ)
Every machine installation requires a three-stage validation process:
- IQ (Installation Qualification): Is the machine installed in accordance with design specifications?
- OQ (Operational Qualification): Does it operate within defined operational ranges?
- PQ (Performance Qualification): Does it deliver consistent results under real production conditions?
4. Electronic Records and 21 CFR Part 11
At FDA-audited facilities, all production data must be stored electronically and an audit trail maintained. This makes SCADA/MES integration mandatory.
5. Precise Filling Tolerance
In pharmaceutical filling machines, the acceptable weight deviation is below ±1 mg. Gravimetric control systems verify the net weight of every ampoule or vial.
